該載體通過自殺基因表達與否篩選陽性重組子。當載體與片段連接成功時，自殺基因無法正確表達，包含重組子的細胞可以正常生長；當載體與片段連接不成功時，自殺基因正確表達，包含載體的細胞無法生長，即“Zero”背景。適用于平端克隆。
